Due to the complementary nature of DNA sequences, once you know the base sequence of the developing/test strand you can automatically work out the sequence of the template strand according to base-pair rules!Adenine pairs with thymine and cytosine pairs with guanine. So if a test strand had the sequence: ATGC then the template strand would have the sequence: TACG.
